Multi-material 3D bioprinting of human stem cells to engineer complex human corneal structures with stroma and epithelium

Bioprinting 2025 Volume 46, Article e00391

Developing cost-effective and scalable multi-material bioprinting technologies that combine multiple cell types is crucial to produce biomimetic, complex human tissue substitutes and overcome the scarcity of transplantable tissues. These technological developments can revolutionize the treatment of several conditions currently dependent on donor tissues, such as corneal blindness. Here, corneal structures consisting of two layers, stroma and epithelium, were manufactured by extrusion-based 3D bioprinting. To take steps towards clinical translation of bioprinting, three clinically compatible hyaluronic acid based bioinks were combined with human adipose tissue and induced pluripotent stem cell derived cell types. Each of the three bioinks was customized to…

Compartmentalized 3D bioprinting of the limbal niche with distinct hPSC-LSC subpopulations for corneal disease modeling

Acta Biomaterialia 2025 Volume 201, Pages 187-197

Limbal epithelial stem cells (LSCs) are essential for corneal epithelium regeneration and visual acuity. The limbal niche’s physicochemical properties regulate LSC function, but their role is not fully understood. Developing in vitro models that mimic the native niche can enhance our understanding of niche functions, despite the challenges of niche complexity. In this study, we created a 3D bioprinted limbal niche model using a hybrid approach that combines two human pluripotent stem cell-derived LSC (hPSC-LSC) subpopulations (p63+ and ABCG2+ cells) within hyaluronic acid (HA)-based bioinks and a stiff polyacrylamide (PA) gel scaffold produced by conventional gel casting. We analyzed the…

FRESH 3D Bioprinting of Alginate – Cellulose – Gelatin Constructs for Soft Tissue Biofabrication

Procedia CIRP 2024 Volume 125, Pages 42-47

The fabrication of three-dimensional (3D) bioprinted free-standing, low viscous, cell-laden hydrogels with good resolution, low cytotoxicity, and mechanical properties, comparable to native soft tissues, is a current challenge in tissue engineering. Recently, a new syringe extrusion approach, called Freeform Reversible Embedding of Suspended Hydrogels (FRESH), has been introduced to enhance 3D-bioprinting of soft hydrogels. Printing is conducted with the material embedded in a thermo-reversible gelatin bath, which acts as supporting material and can also initiate in-situ crosslinking when proper crosslinker agents are added. This work is the first to develop a 3D FRESH printable, low-cost, polymeric hydrogel composed of sodium…

Human Induced Pluripotent Stem Cell-Derived Neural Progenitor Cells Produce Distinct Neural 3D In Vitro Models Depending on Alginate/Gellan Gum/Laminin Hydrogel Blend Properties

Advanced Healthcare Materials 2021 Volume 10, Issue 16, Article 2100131

Stable and predictive neural cell culture models are a necessary premise for many research fields. However, conventional 2D models lack 3D cell-material/-cell interactions and hence do not reflect the complexity of the in vivo situation properly. Here two alginate/gellan gum/laminin (ALG/GG/LAM) hydrogel blends are presented for the fabrication of human induced pluripotent stem cell (hiPSC)-based 3D neural models. For hydrogel embedding, hiPSC-derived neural progenitor cells (hiNPCs) are used either directly or after 3D neural pre-differentiation. It is shown that stiffness and stress relaxation of the gel blends, as well as the cell differentiation strategy influence 3D model development. The embedded…
